ध्वनि-प्रभाव संकेत रेडियो नाटक के लिए अत्यंत महत्वपूर्ण हैं । ध्वनि प्रभाव संवादों के लिए आधार स्तंभ की भांति हैं। संवादों की भावनाओं की रसात्मकता ध्वनि संकेतों से बढ़ सकती है। संवादों में उत्साह, आनंद, दुःख, व्यंग्य, लयात्मकता, उतार-चढ़ाव आदि के लिए संकेत होते हैं। संवाद-संकेत संवादों की जीवंतता, सार्थकता और संदर्भयुक्तता के लिए अपेक्षित होते हैं। संवाद के अंतर्गत पात्र का हँसना, दुःख प्रकट करना, रोना, दीर्घ सांस लेना जैसे संदर्भों का संकेत देने के लिए संवाद- संकेतों की जरूरत पड़ती है। वास्तव में रेडियो केवल श्रव्य- माध्यम होने के कारण इन ध्वनि प्रभाव संकेतों के बिना दृश्यावली स्थापित करने में कठिनाई होती है। उदाहरण के लिए नाटक की घटना में पात्रों को किसी रेलवे स्टेशन में संवाद करते हुए पाया जाता है तो इसके लिए रेल के आने-जाने, उद्घोषणा, यात्रियों और चाय-नाश्ता बेचनेवालों का शोरगुल आदि दृश्य का सृजन ध्वनि प्रभाव संकेतों से ही संभव है। और इसी प्रकार के दृश्यों के लिए कई प्रकार की आवाज़ें यथा दरवाजा खुलने, बंद होने, गाड़ियों के आने जाने, किसी के आने-जाने की आहटें, बच्चों के खेलने की आवाज़ें आदि का सृजन इससे संभव है। आमतौर पर नाटक के दौरान उत्पादित ध्वनियाँ, कुछ पूर्व- रिकार्डेड ध्वनियाँ होती हैं। सामान्यतः रेडियो नाटक का रिकार्डिंग स्टूडियो पर होता है, अतः सभी प्रकार की ध्वनियाँ, ध्वनि प्रभाव प्रत्यक्षतः नाटक के रिकार्डिंग के दौरान ही उत्पन्न करने में कठिनाई होती है, अतः पूर्व में रिकार्ड की गई ध्वनियों का इस्तेमाल किया जाता है।
